sudo dscacheutil -flushcache

or

sudo killall -HUP mDNSResponder

In 10.6 the handling of Unicast DNS is currently taken care of by mDNSResponder - dscacheutil -flushcache currently simply informs mDNSResponder to reactivate (the 2nd command over).
